Title: Transparent Background in Realtime Render

Hi, there..
I always prefer rendering the screenshot. Can you do that with a transparent background?

I don't think so. For a transparent background you need to actually go through the steps of rendering to one of the formats that supports transparent backgrounds. And of course, don't forget to check the "Include Alpha (Transparency)" box when you do.
